What is the CELENA X and what does it do?
The CELENA X is a fully automated high-content imaging system designed for rapid, quantitative cell imaging and analysis. It performs automated plate and slide scanning with four-channel fluorescence, brightfield, phase contrast, and colour brightfield imaging modes. It is used in drug discovery and cell biology to measure cellular phenotypes objectively and reproducibly across multi-well plates (6 to 1536 wells), slides, petri dishes, and culture flasks.
What fluorescence channels and filter cubes are available on the CELENA X?
The CELENA X supports four-channel fluorescence imaging using interchangeable LED filter cubes with a 50,000+ hour lifespan and adjustable intensity. Available filter cubes include DAPI, EGFP, RFP, mCherry, ECFP, EYFP, DSRed, Cy5, Cy7, Cy3/TRITC Long Pass, GFP Long Pass, and Cy5 Long Pass, plus custom filter options. The system is compatible with Olympus, Zeiss, and Logos Biosystems objectives across a magnification range of 1.25x to 100x.
How does the CELENA X autofocus system work and why does it matter for live-cell imaging?
The CELENA X offers both standard image-based autofocus and an optional laser autofocus module. The laser autofocus provides rapid and reproducible focusing while minimising phototoxicity and photobleaching — critical considerations for time-lapse live-cell experiments. The motorised XYZ stage (120 mm x 80 mm X/Y, 10 mm Z travel) combined with the laser autofocus enables consistent focus across large multi-well plates.
Can the CELENA X maintain physiological conditions during live-cell imaging?
Yes. The CELENA X supports optional CX Stage Top Incubators (Basic and Pro models) that provide temperature control, CO2, and CO2/O2 gas supply for maintaining physiological or custom environmental conditions during imaging. This enables long-duration time-lapse experiments, kinetic assays, and other live-cell studies requiring precise environmental control without removing cells from the microscope stage.
What analysis capabilities does the CELENA X software provide?
The CELENA X includes two software components: CELENA X Explorer for protocol setup and seamless integration of imaging and data analysis, and CELENA X Cell Analyser for creating customisable analysis pipelines. Users can identify cellular and subcellular objects, process images for optimal data collection, and perform quantitative measurements. Pipelines can be saved and reused, supporting batch processing from simple fixed-cell assays to complex time-lapse live-cell experiments.
What cell biology assays can be run on the CELENA X?
The CELENA X supports multiple documented application workflows including cell counting, cytotoxicity (brightfield and fluorescence), transfection efficiency, wound healing, apoptosis, confluency monitoring, calcium signaling, phagocytosis, cell cycle analysis, image stitching, and 3D cell model imaging (organoids and spheroids). Both fixed-cell and kinetic live-cell assay formats are supported.
