Zetasizer µV as SEC detector

The Zetasizer µV provides rapid characterization of very small quantities of proteins. Utilising the highly sensitive Single Mode Fibre (SMF) technology enables measurement of dilute solutions of small proteins, detecting down to a hydrodynamic radius of 0.3 nm. Only 2µl. of protein solution is required.

The Zetasizer µV incorporates both Dynamic and Static light scattering. Dynamic light scattering provides accurate determination of molecular size and weight as well as detecting the presence and proportion of aggregates. Static light scattering enables the determination of absolute molecular weight and 2nd virial coefficient - a value indicating how well the protein is likely to stay in solution.

Hardware features

Intelligent software

The software guides users in developing standard operating procedures (SOPs) for repeatable measurements. It incorporates protein specific reports to assist with the interpretation of data. Any measured parameter can be plotted against any other measured parameter eg time vs. temperature.

A set of utilities includes a molecular weight plotter, estimated molecular weight calculator, shape estimate calculator, concentration calculator and scattering functions plotter.

ZETASIZER µV AS SEC DETECTOR

The Zetasizer μV can be used as a flow mode chromatography detector and as a batch mode particle sizer, making it an ideal tool for protein applications in a single package. The Zetasizer μV uses dynamic light scattering (DLS) to measure molecule size and static light scattering (SLS) to determine molecular weight.

The Zetasizer μV is available as part of a complete chromatography system or can be added to an existing Size Exclusion Chromatography (SEC) system. Applications in stand-alone batch mode include the study of molecular size and formulation stability, and in SEC detector mode the measurement of molecular weight, determination of oligomeric state and detection and quantification of oligomers and aggregates.

Malvern's OmniFACE software provides the necessary connection that enables the Zetasizer μV to be linked into any third party SEC system and detectors, so that output signals can be used by the OmniSEC software for results calculation. Connecting the Zetasizer μV is simplicity itself. Using standard chromatography tubing it can be added in series with other detectors such as UV and RI.

The absolute nature of the measurement means that no column calibration is required.

The laser stability and optical setup of the Zetasizer μV ensure excellent baseline stability and signal-to-noise for maximum sensitivity, while the 8 μL flow cell minimizes band broadening.
